Abstract
Objective To investigate the role of resveratrol in the gene expression of T-bet/GATA-3 in Jurkat T cells,and to explore the mechanisms underlying this sensitizing effect of the change of TH cell subpopulation group. Methods The related cytokines about Thl/Th2 were detected by ELISA and T-bet and GATA-3 mRNA were detected by RT-PCR after Jurcat T cell are cultured with different doses of Resveratrol for 48 hours. Results Resveratrol inhibited the expressions of mRNA of T-bet and GATA-3 ( F = 6308, P < 0. 05; F = 3262,P< 0.05) and the secretions of the related cytokines about Thl/Th2(F = 2604,P <0. 05; F =2465, P<0. 05). And these two effects are all concentration related. Conclusion Resveratrol could inhibit Th0 cells to differentiate to Thl cells by down-regulating transcription factor T-bet. Resveratrol also could inhibit Th2 cells differentiation from decreasing the expression of mRNA of GATA-3. And these two effects are all concentration related.
