Abstract

Scanning electron microscopy can be used to observe the crystal and spore produced by Bt strain during sporulation, which is recognized as the fastest and the most intuitive method for identifying Bt strain. This protocol is a routine method in our laboratory used to observe morphology of Bt crystal protein and spore by the means of scanning electron microscope.

  • Bt 最主要特征就是在生长后期产生芽孢和伴 孢晶体,因此在这个时期用扫描电子显微镜来观 察其形态是最佳的。本案例中,我们以 Bt 分离株 S3299-1 (Fang et al, 2011)为实验材料,锇酸固定 法对生长后期的 Bt 菌进行扫描电镜鉴定。如图 2 结果显示,在 Scanning electron microscope (SEM) HV 30.0 kV View field: 7.223 μm WD: 7.313 mm 30.00 kx 观察条件下,能清晰 观察到 S3299-1 菌株的芽孢以及产生的菱形晶体。
