Abstract
1. 1. A nitrate reductase (NADH:nitrate oxidoreductase, EC 1.6.6.1), which catalyzes the reduction of nitrate to nitrite by NADH, has been found to be present in fresh extracts of nitrate-grown Chlorella pyrenoidosa in the form of a precursor of low activity. The enzyme precursor can be activated in vitro over 100-fold in a slow process which is accelerated by added nitrate and phosphate buffer of low pH. 2. 2. A partial purification of nitrate reductase is described. The enzyme is activated during the purification process itself. The partially purified enzyme contains a cytochrome, which is similar to the cytochrome associated with the assimilatory nitrate reductase of Neurospora, studied by A. Nason and his associates.
