Abstract

ABSTRACTIt is a quality assurance/quality control requirement to perform internal validation studies on new or modified reagents used in forensic DNA typing, even when performance verification studies have been conducted by the manufacturer. In 2006, Applied Biosystems (AB) released modified AmpFlSTR® Profiler Plus™ and AmpFlSTR® COfiler™ allelic ladders. Prior to adopting the modified allelic ladders, an in-house validation study was conducted and it was found that there was no significant difference in sizing precision and genotyping concordance between the previous and modified allelic ladders. A definite improvement in allele balance was observed across all loci with both allelic ladders. However, we did not necessarily observe an overall increase in peak heights as claimed by the manufacturer. We can conclude that the modified allelic ladders are still robust, reliable, and reproducible for forensic DNA typing.
