Abstract

In vitro methods to predict the adequacy of stored marrow would be clinically valuable. We investigated the CFU-C assay to predict for hematopoietic recovery after high-dose therapy. In acute leukemia (AL) marrow was collected in first remission and in solid tumor (ST) after failure of previous therapy. Marrow was stored in liquid nitrogen at –192°C. Before transplant (TR), patients with AL (n = 11) received piperazinedione 50 mg/sq m and total body irradiation 850 rads. Patients with ST (n = 13) received Cytoxan 6 g/sq m, VP-16 500 mg/sq m, and BCNU 300 mg/sq m. Marrow was then thawed, and the number of CFU-C per 10s cells infused determined. In patients with ST, hematopoietic recovery after TR was not related to CFU-C content in the patient’s marrow before chemotherapy, but to the total number of CFU-C or CFU-C/kg infused.
