Abstract

Objective To investigate the molecular characteristics of noroviruses among viral diarrhea cases in Baoan district,Shenzhen.Methods From January 2010 to December 2011,486 stool samples were collected from suspected viral diarrhea patients in Baoan district,Shenzhen.Nucleic acid of GⅡ noroviruses were detected using real-time polymerase chain reaction (real-time PCR).The positive samples for GⅡ norovirus were randomly selected to perform reverse transcription-polymerase chain reaction (RTPCR) with the primers COG2F/G2-SKR,while all the negative samples for GⅡ norovirus were performed RT-PCR with the primers GI-SKF/GI-SKR.Positive PCR products were purified and sequenced,and the sequences were analyzed by Clustal W and MEGA 5.0 programs,then phylogenetic tree was constructed and genotype was identified.Results Among 48 positive samples for GⅡ norovirus,the numbers of G Ⅱ.4/ 2006b,G Ⅱ.4/2008 and GⅡ.5 were 36,3,and 9 respectively; No GⅠ norovirus was found.Conclusions From January 2010 to December 2011,G Ⅱ was the most important genotype of norovirus,and G Ⅱ.4/2006b variant was identified as the predominant strain in Baoan district,Shenzhen.
