Abstract

Objective To investigate the effects of Cisplatin(CDDP) and radiation on telomerase and cell cycle of human laryngeal squamous cells and explore the mechanism.Methods Hep 2 human laryngeal squamouscells were exposed to CDDP or 60 Co gamma radiation.FCM analysis,PCR based Telomeric Repeat Amplification Protocol(TRAP) coupled with ELISA and polyacrylamide gel eletrophoresis (PAGE)were used to estimate the change of telomerase activity and cell cycle. Results Through exposure to CDDP or irradiation,significant increase of telomerase activity was observed,on a dose dependent manner.Analysis of cell cycle indicated increase of S phase cell percentage induced by CDDP and of G2/M phase cell percentage by irradiation.The increase of telomerase activity induced by therapy was correlated with the increase of S phase cell percentage (r=0.5371,b=7.8393).Conclusion The change of telomerase activity induced by therapy were correlated with the change of cell cycle.
