Abstract

1. 1. Phosphoglycerate, pyruvate, and hexose diphosphate increased oxygen production from illuminated spinach juice, but gave at most 16% of the stoichiometric yield calculated on the basis of the amount of substrate added. 2. 2. Evidence is presented for the occurrence in spinach juice of a substance which, in the presence of illuminated chloroplasts and catalytic amounts of TPN, produces oxygen. A specific coupling with the fixation of carbon dioxide could not be proved in juice preparations. 3. 3. The direct reduction of TPN by chloroplasts in a light reaction was demonstrated with the aid of the malic enzyme system. Under these conditions oxygen evolution occurs coupled with the fixation of carbon dioxide, as shown by the radioactive tracer method.
