Abstract

Nanoparticles-protein interactions have gained a lot of attentions in several fields including nano-bio safety, nano-bioscience, nano-biomedicine, and nano-biotechnology. Hence, in this study the effects of Fe3O4@Arg (RMNPs) on the lysozyme structure, refolding, and denaturation along with its electrostatically effect on amyloid aggregation of lysozyme have been investigated. RMNPs were synthesized based on the Massart method and XRD, SEM, FT-IR, VSM, and Zeta Potential (DLS) were applied to investigate its structural and morphological properties. In addition, ThT assay and SEM were utilized in order to monitoring the inhibiting activity of RMNPs and its effects on the amyloid fibrils. Kosmotropic and chaotropic effects of RMNPs on lysozyme are investigated at lower and higher RMNPs/protein ratios, respectively. The result of this study showed that the protein folding, helicity, and half-life are improved at a low concentration of RMNPs. RMNPs improved depolymerization and disassembly of aggregates by reducing the aggregation and destroying fibrils.
