Abstract

Over half of the genetic predisposition to autoimmune insulin-dependent diabetes mellitus (IDDM) is determined by one or more genes within the major histocompatibility complex (MHC) (1). Serologic, cellular, and DNA (restriction fragment length polymorphisms; RFLP) HLA typing methods have previously suggested that an IDDM susceptibility is closely linked to the HLA-DQ subregion (2). By direct sequencing of HLA genes from IDDM patients, we have established a correlation between a polymorphic residue of the DQB1-chain (position 57) and IDDM susceptibility/resistance that indicates a direct role for the DQ molecule in disease susceptibility (3).
