Abstract

Using Sephadex LH-20 column chromatography and solvent systems composed of cyclohexane and ethanol, a Chromatographic method is reported for the separation of free steroids in biological material. These systems are in routine use for the purification and separation of plasma corticosteroids prior to competitive protein binding analysis. The systems are also used for group separation of urinary steroids before gas Chromatographic analysis. Routine measurements of aetiocholanolone and pregnanetriol in urine by gas chromatography are carried out in this laboratory following Sephadex LH-20 chromatography. These systems have been particularly useful in the ‘profile’ analysis of urinary steroid metabolites in patients with endocrine disorders.
