Abstract

Fat storage‐Inducing Transmembrane proteins 1 & 2 (FITM1/FIT1 & FITM2/FIT2) are an evolutionarily conserved family of endoplasmic reticulum resident membrane proteins that play an important role in the formation of triglyceride lipid droplets (LDs). FIT1 and FIT2 have 6 transmembrane domains with their N‐ and C‐termini facing the cytosol. However, FIT proteins do not have similarities to known protein domains, making it difficult to infer mechanism based on primary sequence. We have utilized both novel in vitro biochemical assays and mouse transgenic and gene‐targeted knockouts of FIT genes to determine function. These studies aimed at understanding the biochemical mechanism and physiological function of FIT proteins will be presented.
