Abstract

Objective To study the expression of Caspase 4(Casp4) gene in Kawasaki disease and its relation with nuclear factor-kappa B (NF-κB) signaling pathway. Methods The blood samples of 15 patients with Kawasaki disease from January 2012 to January 2015 in Shanghai No.6 People's hospital East hospital were collected and the peripheral blood mononuclear cells were extracted.The expression of Casp4 gene and NF-κB signaling pathway related factors were detected by mRNA quantitative detection.The expression of Casp4 protein and NF-κB signaling pathway related factors were detected by enzyme-linked immunosorbent assay(ELISA) and Western blotting. Results The results of mRNA quantitative detection, ELISA and Western blotting showed that the expression of Casp4 gene in patient with kawasaki disease was significantly higher than that of healthy controls.NF-κB signaling pathway related factor expression also changed significantly.Data analysis showed that there were a linear relationship between the NF-κB p65 and Casp4 gene mRNA expression in patients with kawasaki disease. Conclusion The NF-κB signaling pathway was activated while infants affected by environmental factors and genetic factors.NF-κB was released into the nucleus, causing the altered expression of Casp4 gene.High expression of Casp4 in patients with kawasaki disease was related to NF-κB signaling pathway.
