Abstract

Adanosine triphosphate(ATP) is the basic energy source for living cells.It rapidly disappears when cells die.So measuring the ATP levels may estimate the viability of cells exposed to drugs.Dose-response curve can be determined in mouse fibroma cell line:L929 treated with chemotherapeutic agents by the ATP-bioluminescence assay and The different in vitro response of 6 ovarian carcinoma (5 from fresh tumor tissues,1 from ascites)treated with etopside,cis-plating,5-fluorouracil and adriamycin can be investigated.The coefficient of variation for triplicate assays ranges from 1.2 to 15.8%.It means high reproducibility of the assay.This presentation also reported that single cell suspension(include 30 cells clusters) can be separated from tissue fragments by means of the enzyme cocktail(collagenase,Dnase,pronase).The viable cells were over 90%.0.5% Argarose/McCoy underlayer can prevent the growth of anchorage-dependent nonmalignant cells.This study demonstrates that ATP bioluminescence assay is a sensitive,efficient method for tumor chemosensitivity test.Correlation of in vitro drug sensitivity and in vivo patient response worth further studying.
