Abstract

Objectice To investigate the change of the expression of intestinal trefoil factor family mRNA and effects of ulinastatin on it in the rat model of sepsis.Mathods Sixty Sprague-Dawley rats were randomly divided into control,sepsis,pretreated and treated groups (n=15). Sepsis was induced in the later three groups by cecal ligation and puncture(CLP). The pretreated groups were received 25 00Ou/Kg ulinastatin at 2 hours before operation and the ulinastatin treated groups were received 50 000u/Kg ulinastatin at 2 hours after operation.Some ileum mucosas were taken at 12h after CLP. The pathological change was observed and the expression of trefoil factor family mRNA was detected with RT-PCR. Results The expression of TFF3 mRNA in sepsis group significantly decreased compared to control one(P<0.05);Pretreated and treated groups significantly increased compared to sepsis group (P<0.05);Pretreated groups significantly increased compared to treated groups (P<0.05).Conclusions The expression of intestinal TFF3 mRNA significantly decreased in sepsis,which could be improved by the treatment of ulinastatin leading to significantly intestinal mucosal protection. The pretreatment may be more effective than the therapeutic treatment in the rat model of sepsis. Key words: Sepsis; Trefoil factor family; Ulinastatin; Cecal ligation and puncture
