Abstract

Objective To investigate the effects of pre-intervention parecoxib sodium on premenstrual syndrome (PMS) model rats.Methods 36 rats were randomly divided into 3 groups according to the results of vaginal secretion smear,the normal group (group C),the model group (group M),and the pre-intervention parecoxib sodium group (group A).PMS models were prepared by chronic restraint stress,which persist for 7 d.Group A was pre-administered parecoxib sodium 0.72 mg/kg from the fifth day to the seventh day.All the rats were sacrificed at the eighth day.Then we applied immunohistochemical to detect the cycloxygenase-2 (COX-2) and prostaglandin E2 (PGE2) expression on rat endometrium,and western blot to detect the COX-2 and PGE2 expression on rat hippocampus.Results ① COX-2 expression on rat endometrium of group M and group A was significantly higher than group C(P<0.01).COX-2 expression on the endometrium of group A(0.31±0.12) was 32% lower than group M(0.46±0.11)(P<0.05).② PGE2 expression on rat endometrium of group M and group A was significantly higher than group C (P<0.01).PGE2 expression on endometrium of group A (0.39±0.09) was 18.9% lower than group M (0.49±0.08)(P<0.05).③ COX-2 expression on hippocampus of group M and group A was significantly higher than group C (P<0.01).COX-2 expression on hippocampus of group A (0.72±0.05) was 20.8% lower than group M (0.91±0.03)(P<0.05).④ PGE2 expression on hippocampus of group M and group A was significantly higher than group C (P<0.01),COX-2 expression on hippocampus of group A (0.92±0.06) was 18% lower than group M (1.13± 0.03) (P<0.05).Conclusions Parecoxib sodium pre-intervention could decrease COX-2 and PGE2 expression on endometrium and hippocampus in premenstrual syndrome model rats.
