Abstract
Objective To study the effect of response gene to complement 32 (RGC32) gene on the proliferation,apoptosis and invasion of glioma cells.Methods 5-ethyny-2' deoxyuridine (EdU) cell proliferation assay was used to detect the effect of RGC32 gene on the glioma cells proliferation; Annexin Ⅴ-fluoresceine isothiocyanate (FITC) apoptosis detection was used to detect the effect of RGC32 gene on the glioma cells apoptosis ; Transwell invasion assay was used to detect the effect of RGC32 gene on the glioma cells invasion.Results Overexpression of RGC32 gene in glioma cells could promote cell proliferation and invasion which.increased by 56.52% and 119.05% respectively (P < 0.05),but had no effect on cell apoptosis (P > 0.05).Inhibition of RGC32 gene in glioma cells could inhibit cell proliferation and invasion which decreased by 43.47% and 65.32% (P < 0.05),and promote cell apoptosis which increased by 128.57% (P < 0.05).Conclusion Our research shows that abnormal expression RGC32 gene may be an important pathogenic mechanism of glioma.By inhibiting the expression of targeted genes RGC32 can effectively promote apoptosis and inhibit glioma proliferation and invasion.
