Abstract
Objective To invetigate The effect of Pygo2 to renal carcinoma cell OS-RC-2 apoptosis in vitro.Methods Lentiviral-mediated reorganization of the three plasmids,to establish the Pygo2 silence group,the Pygo2 overexpression group and the corresponding control group.Lentiviral was used to infect the renal carcinoma OS-RC-2 cells,and the Pygo2 shRNA infection efficiency was detected by realtime quantitative polymerase chain reaction (Real-time PCR).Propidium iodide (PI) staining to detect cell apoptosis and detecting apoptosis protein C-poly adenosine diphosphate-ribose polymerase (PARP) expression using Western blotting.Results Pygo2 was inhibited 80%.Rate of apoptosis were 6.52%,9.01%,61.09%,6.88% in Pygo2OE,GFP,Pygo2 shRNA,Pygo2 scrRNA.Inhibition of Pygo2 expression,apoptosis peaks was prominent.The down-regulation of Pygo2 can promote the generation of c-PARP.Only pygo2shRNA expressed c-PARP by Western blotting.Conclusion The silence of Pygo2 gene was able to promote apoptosis of renal cell carcinoma OS-RC-2 in vitro.
