Abstract

Binding and acceptor site activity of intrinsic factor from depepsinized gastric juice was studied after exposure to a wide range of pH. Binding site activity remained stable when preincubated at pH 1.5 to 9.0 for 2 hours whereas acceptor site activity remained stable when preincubated for 4 hours over a similar pH range. A modest reduction in binding activity occurred at pH 10 during the first hour and at pH 1.5 and 2.5 during the third and fourth hours of preincubation. These results were interpreted as demonstrating the relative stability of the binding and acceptor sites of intrinsic factor over a wide range of pH.
