Abstract

Folate is crucial for DNA synthesis,repair,and DNA methylation.Folate deficiency induces genomic instability such as DNA strand breakage,chromosomal structural aberration,DNA methylation profile and expression pattern changes.Folate receptor is a high-affinity folate binding protein.The expression of different folate receptor isforms is tissue specific.The isoforms differ expression and function in normal and malignant human tissues.Therefore,it has been studied extensively as a tumor cellular surface marker for targeted drug delivery.The absorbing of folate is effected by the number and isform of folate receptor,meanwhile,folate conjugates target is expected to be competitively inhibited by excess free folate present in the plasma.This paper reviews the relationships between folate metabolism and folate receptor,and its applications in the therapy of tumor.
