Abstract

1. Dibutyryl cyclic AMP enhanced rhythmic contractions and increased resting tension of rat ileal smooth muscle, but did not alter tension in the non-spontaneous smooth muscle of the vas deferens. 2. This agent slightly enhanced the tension of pre-existing tonic KCl contractures, while pre-treatment with dibutyryl cyclic AMP also potentiated the phasic part of KCl-induced contractures. 3. Dibutyryl cyclic AMP was without significant effect on calcium uptake by microsomes isolated from ileal smooth muscle but it inhibited calcium uptake by isolated mitochondria. 4. Although dibutyryl cyclic AMP was without immediate effect on Ca 45 efflux from either the ileum or vas deferens, a large protracted stimulation of efflux occurred after return to control saline in both preparations. 5. It is concluded that in these two smooth muscles, cyclic AMP does not seem to be involved in the relaxation mechanism.
