Abstract

Objective: To study the effects of curcumol on the protein expression of JAK2 and STAT3 in SKOV3 and to investigate its treatment on molecular mechanism of ovarian cancer. Methods: Choose curcumol of different concentrations to act on human ovarian cancer cell line SKOV3, and extract the corresponding cell protein, and detect the protein expression of JAK2 and STAT3 by western blotting. Results: The protein expression of JAK2 and STAT3 in SKOV3 are significantly inhabited by curcumol, and its strength will enhance with the increase in drug concentration, and it shows in a dose-dependent manner. Conclusion: Curcumol can significantly inhabit the proliferation of SKOV3 cells, and induce apoptosis, and achieve its mechanism by regulating the protein expression of JAK2 and STAT3.

Curcumol; Human ovarian cancer cell line SKOV3; J AK2/STAT3 Signaling;Proliferation 1 黑龙江中医药大学附属第一医院,黑龙江哈尔滨 150040 2 黑龙江省大庆油田总院集团油田总医院中医科,黑龙江大庆 163001 3 黑龙江中医药大学(在读研究生),黑龙江哈尔滨 150040 【摘要】 目的 研究莪术醇对 SKOV3 细胞中 JAK2 和 STAT3 蛋白表达的影响,进而探讨莪术醇治疗卵巢癌的分子机制。方 法 选择不同浓度莪术醇作用于人卵巢癌细胞株 SKOV3,提取相应细胞蛋白,利用蛋白印迹法测定 JAK2 和 STAT3 蛋白的表 达。结果 莪术醇对卵巢癌 SKOV3 细胞中 JAK2 和 STAT3 的蛋白表达有明显的抑制作用, 其抑制强度随药物浓度的增加而增 强,呈剂量依赖性。结论 莪术醇能明显抑制 SKOV3 细胞的体外增殖,诱导细胞凋亡,其机制可能是通过降调节 JAK2、STAT3 蛋白表达来实现的。 目前,卵巢癌是女性最常见的生殖肿瘤之一,其致死率居各类妇科肿瘤之首。中药莪术(姜黄根茎)具 有破血行气、消积止痛之功效,应用于癥瘕积聚、经闭、心腹瘀痛等疾病。《药品化义》中记载:“莪术 味辛性烈,专攻气中之血,主破积消坚,去积聚痞块,经闭血瘀,扑损疼痛。”莪术中起主要作用的为挥 发油类物质,包括莪术醇、莪术二酮、吉马酮等。莪术醇是一种愈创烯型倍半萜类的半缩酮,具有抗肿瘤、 抗增殖、抗炎、抗肝纤维化、抗氧化、低细胞毒性灭菌等特性[1,2,3,4]。现代研究表明莪术挥发油制剂对多种癌 细胞有直接破坏作用,并能通过免疫系统使特异免疫增强而获得明显的免疫保护效应,从而具有抗癌作用。 莪术醇在体外能明显抑制 MCF7、MM231、HeLa 细胞和 OVUL -2 等妇科肿瘤细胞系的增殖和 RNA 合成[5]。 然而目前,对莪术诱导细胞凋亡的特点和分子机制却不甚了解。
