Abstract

Objective To observe the time-course change in expression of inducible nitric oxide synthase(iNOS) and tumor necrosis factor-α(TNF-α) in the liver of mice infected with Toxocara canis. Methods The livers of 16 healthy Kunming female mice were collected uninfected or infected with T. canis on day 3, 5, 9 post infection( p.i.) , totle RNA of livers were extracted with RNAiso reagent, then time-course change of the mRNA expression of iNOS and TNF-α was detected by real time fluorescent quantitative RT-PCR.Results There was no expression of iNOS in the livers of mice uninfected. The mRNA expression of iNOS increased significantly and reached peak at day 3 p. i. , then the expression decreased gradually on the day 5 and 9 p. i.. The mRNA expression of TNF-α was detectable in the livers of both infected and uninfected mice,and the pattern of time-course change was same as that of iNOS. Conclusion The infection of T. canis can induce coexpression of iNOS and TNF-α in the liver of mice, both iNOS and TNF-α had the same pattern of time-course change of expression.
