Abstract

The availability of winegrapes with a higher freezing tolerance than currently used V. vinifera cultivars is desirable for the Wine Industry in Canada. One way to accomplish this is by crossing V. riparia with V. vinifera. This study reports on the analysis of techniques that might be used in such a program. Electrolyte leakage values obtained from grapes either before or after a low temperature treatment did not show a significant difference between the two species. Arabidopsis DNA extracted by a quick ethanol-based method could be amplified but this was not the case for grape DNA. PCR primers were developed to identify different alleles of Vitis CBF1 and Vitis CBF4. Analyses of 2 V. vinifera cultivars, 17 V. riparia accessions and 6 winegrapes from Italy and Germany showed that these primers could be useful for marker assisted selection.
