Abstract

Oriented circular dichroism (oCD) spectroscopy is a challenging but revealing technique that is used to ascertain the tilt angle of membrane proteins in oriented lipid bilayers. The preparation of conventional oCD samples, where lipid bilayers are deposited on a support substrate, is a laborious process which can require extensive repetition and optimisation to produce well aligned bilayers with good optical properties. Even then uncertainty over the depth of bilayers deposited and effective path-length results in inconsistency between samples and also within different regions of the same sample.
