Abstract
A staining method for β-alanine, based on the treatment of high voltage paper electrograms or paper chromatograms with ninhydrin and sodium carbonate, is described. The electrogram or chromatogram after drying is stained with a solution of 0.4% ninhydrin in water-saturated n -butanol. The ninhydrin colour is developed by heating and the β-alanine colour formed by dipping the paper strip into a 2% sodium carbonate solution. The dilute alkali treatment shifts the absorption maximum of the ninhydrin β-alanine complex from 570 nm to 600nm and a characteristic blue colour is formed. This reaction, which is similar to one described earlier for phenylalanine, is applicable to studies of aminoacidurias and would be especially helpful in detecting cases of hyper-β-alaninemia. The sensitivity of the method is similar to that of the β-alanine ninhydrin reaction.
