Abstract

Bacteria that carry out N,N,N',N'-tetramethyl-p-phenylenediamine (TMPD) oxidations are “oxidase-positive” species, which possess a membrane-bound cytochrome-dependent terminal oxidase. The components needed for TMPD oxidation are c-type cytochrome(s) integrated with a terminal oxidase component, like cytochrome o or a + a3. The obligate aerobe, Azotobacter vinelandii, has the highest cellular respiratory rate of any organism known and for this reason its cytochrome oxidase is of particular interest. This high oxidative rate is also reflected by the terminal TMPD oxidase activity, which concentrates exclusively in the electron transport particle (ETP). A 20-fold purification of the Azotobacter TMPD oxidase enzyme complex, by a detergent solubilization procedures, shows it to be an integrated cytochrome c4 + c5: cytochrome o complex.
