Abstract

1. 1. Paper electrophoresis affords a method of complete separation of two enzymes from crude elastase preparations. 2. 2. It is shown that one of these enzymes (E 1), which is without the power of causing any protein to pass into solution when tested alone, activates the other, E 2, when added simultaneously. 3. 3. The ratio of polysaccharide to protein liberated by a mixture which is mainly E 1 is far greater than that liberated by a mixture which is mainly E 2. This is taken to indicate that E 1 reacts in some way with the mucoid fraction of the elastin fiber. 4. 4. Use is made of fractional ammonium sulfate precipitation and dialysis to obtain enzyme preparations which are rich in either of the two enzymes. 5. 5. Similar fractionation is reported after adsorption on various solid adsorbents including the substrate itself, which removes E 1 completely from the preparations.
