Abstract

The liver is responsible for the net excretion of cholesterol from the body through biliary excretion of bile acids and free cholesterol. The successful cloning of the rate-determining enzyme in bile acid synthesis, cholesterol 7α-hydroxylase, has made study of the molecular regulation of bile acid biosynthesis and its relation to lipoprotein metabolism possible. With a detailed understanding of the adaptive mechanisms of hepatic cholesterol metabolism we can explain the mechanisms for lipid-lowering therapy in man.
