Abstract

ISEE-550 Objective: Our aim was to assess whether the functional Val16Ala polymorphism in the manganese superoxide dismutase (MnSOD) gene is associated with the risk of oral squamous cell carcinoma (OSCC). Materials and Methods: Participants with OSCC or oral precancerous lesions (OPL) were recruited from Kaohsiung Medical University hospital with physician diagnosis and histological confirmation. Healthy controls were randomly selected from a community survey for oral cancer. The MnSOD genotypes were determined using the PCR–RFLP technique. Detailed information including substance usage (cigarette smoking, betel quid chewing, and alcohol drinking) for each participant was obtained from standardized questionnaire. We used logistic regression to model the association between the genotype and OSCC risk. Results: Two hundred and twenty-seven OSCC patients, 108 OPL patients, and 254 healthy controls were recruited into the present study. Habits of each substance usage were risk factors for OSCC. Compared with Ala allele carriers, the risk of Val/Val genotype for OSCC transformation from OPL was 0.41 (95% CI=0.23–0.68) and risk for OSCC development was 0.50 (95% CI=0.34–0.74). Stratified by 3 kinds of substances' usage or not, the reduced risks of MnSOD Val/Val genotype on the OSCC development were increased 2-fold in participants with cigarette smoking habit or those without the habit of alcohol consumption. After adjusted with the possible confounding factors, the MnSOD Val/Val genotype was associated with the OSCC development (OR=0.34, 95% CI=0.18–0.63) or OSCC transformation from OPL (OR=0.42, 95% CI=0.24–0.72). Conclusions: We suggest that MnSOD Val/Val genotype can reduce the OSCC risks. Gene and environmental interactions are found between MnSOD genotypes and substance usage.
