Abstract

Objective To determine the aqueous concentrations of interleukin 6(IL-6)and vascular endothelium growth factor(VEGF)in eyes with silicone oil tamponade and proliferative vitreoretinopathy(PVR).Methods Thirty-nine patients(40 eyes)underwent vitrectomy at Peking University Eye Center were enrolled in the study prospectively and aqueous samples were obtained from these vitrectomized eyes,14 patients(15 eyes)with silicone oil tamponade,15patients(10 eyes)with PVR and 10 patients(10 eyes)with idiopathic macular hole and epi-macular membrane.The silicone oil filled eyes were also divided into subgroup 1(with recurrent membranes,6 eyes)and subgroup 2(without recurrent membranes,9 eyes)according to the presence of recurrent epiretinal or subretinal membranes.The undiluted aqueous samples were collected by paracentesis and the levels of IL-6 and VEGF were analyzed by ELISA.Results The aqueous level of IL-6 in the silicone oil tamponaded eyes(307.33±152.68 pg/ml)was significantly higher than PVR(235.35±98.41 pg/ml)and idiopathic macular hole and epi-macular membrane eyes(30.18±14.3 pg/ml).The aqueous level of IL-6 in the eyes of silicone oil subgroup 1(408.57±151.21 pg/ml)was significantly higher than subgroup 2(239.84±117.02 pg/ml)and PVR eyes.There were no significant differences in the aqueous level of VEGF between silicone oil filled eyes(294.94±121.81 pg/ml)and PVR eyes(297.43±134.06 pg/ml),but both were significantly higher than the eyes with idiopathic macular hole and epi-macular membrane(32.78±12.9 pg/ml).Conclusions The aqueous levels of VEGF and IL-6 in silicone oil filled eyes are significantly higher than PVR eyes.The accumulation of proliferative cytokines such as VEGF and IL-6 in silicone oil filled eyes can stimulate the recurrence of PVR and lead to the failure of retinal detachment surgery.
