Abstract

This study demonstratedthat pretreated with Evans blue could prevent the compound 48/80 (C48/80) induced allergic circulatory shock and systemic inflammation.Different groups of Sprague‐Dawley rats received i.v. injection of a dose of Evans blue (0, 5, 10, or 50 mg/kg) just 20 sec prior to i.v. injection of C48/80 (200 μg/kg, over 2 min) that was followed by Evans blue to total dose 50 mg/kg as the tracer dye. The results of study found that pretreatment with Evans blue in a dose of 10 or 50 mg/kg could inhibit C48/80‐induced sudden drops in mean arterial and pulse pressures, and pretreatment with Evans blue in a dose of 5, 10, or 50 mg/kg inhibited C48/80‐induced extensive plasmaextravasation, mast cell degranulation, and edema formation in various organs including the airways, esophagus, and skin. In conclusion, Evans blue pretreatment prevented rats from C48/80‐triggered allergic shock and systemic inflammation, possibly mainly due to inhibition of mast cell degranulation.
