Abstract

The stimulation of interleukin-1β (IL-1β) in carp, Cyprinus carpio, injected with peptidoglycan (PG) was investigated by enzyme-linked immunosorbent assay (ELISA) and RT-PCR. Stimulation of IL-1β at significant levels was first recorded at 4 h and was at its maximum 8 days after PG injection. Using RT-PCR, the IL-1β genes in kidney cells of carp treated with PG was also expressed. The PG-injected carp serum costimulated the mitogenic responses of carp lymphocytes by phytohaemoagglutinin (PHA-P). Furthermore, the superoxide anion production of kidney phagocytic cells in carp injected with PG increased significantly. These results suggest the production of IL-1β was due to the immune stimulation by PG.
