Abstract

Experiments were performed to determine whether the action of ICSH in stimulating incorporation of ffiP into testicular phospholipids of hypophysectomized rats was exerted upon both Leydig cells and seminiferous tubules or only upon Leydig cells. The germ cells of intact rats were destroyed by X-irradiation, leaving Leydig cells and Sertoli cells. Leydig cells were dissected from tubules and KP measured in phospholipids of Leydig cells and tubules 5 hr after a single intraperitoneal injection of ICSH. The number of Leydig cells in the tubule fractions after dissection was counted. Leydig cells clearly showed greater incorporation of 32P after ICSH than in controls and the increase in incorporation seen in the tubule fractions was considerably reduced by correcting for Leydig cell contamination. The small response seen after such correction may represent a true response of tubules to ICSH or may result from underestimation of Leydig cell contamination in tubule fractions.
