Abstract

Ten nitrosoguanidine-induced temperature-sensitive ( ts) mutants of the toxinogenic temperate corynebacteriophage β have been isolated. They have been assigned to eight complementation groups on the basis of spot tests on plates and mixed infection complementation tests in liquid culture. A linear map of the ts markers, the diphtheria toxin structural gene marker crm 30, and the markers c, h, and h′, was constructed by two and three factor crosses. The toxin structural gene tox mapped near the c marker, between two clusters of ts loci. Other three factor crosses established colinearity with the map of certain ts mutants described by Matsuda et al. (1971) . A model is proposed to explain aberrant results obtained in crosses involving superinfection of uv-induced lysogenic cells.
