Abstract
<h3>Objectives</h3> Cancer is one of the major public health problems worldwide, representing the leading cause of morbidity and mortality. The National Institutes of Health has recorded that around 40,000 publications related to cancer are published every year; 99% are claims and very few are supported by evidence sufficient for regulatory approval. To assist in overcoming this problem, we need to establish more qualified protocols and update novel theories in research methodologies. Our aim was to help in establishment the most approached outline of research design in using the cell line carcinomas and their pharmacogenomic applications. <h3>Study Design</h3> We investigated the effects of hydroxygenkwanin (HGK), a traditional Chinese medicine, against oral squamous cell carcinoma chromosomal instabilities using different designs. SCC4, SCC9, SCC15, and SCC25 were treated with dissolved HGK. Expression was analyzed and cells were stained by Propidium Iodide (PI) for ploidy and analyzed using FACScaliber. For unbiased, we used 2 logically protocols in analyzing and comparing the results. siRNA and transfected Ca9-22 cells were used as control. <h3>Results</h3> This study found that cells treated with HGK exhibited significantly decreased chromosomal instabilities channels compared with nontreated HGK-overexpressing cells. We recorded that the number of hyperdiploid passages was decreased but there were wide differences between the previous results and others when using the second control. To confirm the results, all passages were investigated before they were treated. We reported that the significances were differences using different generations and different controls. We confirmed that cell lines already have uncontrolled multichromosomal instabilities based on cryopreservation. <h3>Conclusions</h3> We have established that cell lines are not a good choice for studying the effect of treatments on genetic alterations.
