Abstract

The precise temporal coordination of the three cardinal oscillations of non-rapid eye movement sleep – thalamic sleep spindles, cortical slow oscillations (SOs) and hippocampal sharp wave ripples – mediates memory consolidation. Patients with schizophrenia have impaired sleep-dependent memory consolidation that correlates with both spindle deficits and with spindle-SO coordination. In this and a previous clinical trial eszopiclone, a non-benzodiazepine sedative hypnotic, increased spindles but failed to improve memory. Here, we tested the hypothesis that this failure reflects that eszopiclone disrupted spindle-SO coordination.
