Abstract

Purpose: All-trans retinoic acid (atRA) is an active metabolite of vitamin A. It controls the transcription of hundreds of genes and has described anti-inflammatory actions in several other systems, including inflammatory arthritis and acne. atRA plays an essential role in embryonic limb development, but its role in the adult cartilage is poorly studied. Hypomorphic variants in ALDH1A2, that encodes for the key enzyme in the synthetic pathway of atRA was identified in a hand osteoarthritis Genome Wide Association Study (GWAS), indicating that this pathway may have a role in OA pathogenesis.
