Abstract

Genetic code expansion (GCE), combined with click chemistry, offers a powerful tool for protein labeling with single-residue precision. This approach is widely suitable for visualizing proteins in both fixed and live cells, and readily compatible with advanced imaging techniques, e.g., superresolution microscopy and fluorescence-lifetime imaging microscopy (FLIM). GCE is achieved by introducing an orthogonal tRNA/synthetase suppressor pair into the living host, to reassign a stop codon to incorporate a noncanonical amino acid (ncAA).
