Abstract

In confluent resting Swiss mouse 3T3 cells, prostaglandin F2α (PGF2α) stimulates the initiation of DNA synthesis and cell division. This effect is enhanced by insulin and reflected by an increase in the rate into S phase (1). Recently it has been demonstrated that PGE1 and PGE2 (at concentrations that do not stimulate DNA synthesis) have a synergistic effect on PGF2α alone, or with insulin (2). In this communication we report that retinoic acid, as well as analogues of retinoic acid, enhanced the effect of PGF2α and insulin. This synergy was not further increased by PGE1 or PGE2. Indomethacin, an inhibitor of prostaglandin synthesis, did not affect the synergy of retinoic acid with PGF2α, indicating that retinoic acid did not extert its effect by increasing prostaglandin synthesis.
