Abstract

Objective To investigate the effect of deguelin combined with cisplatin in human gastric cancer cell line MGC-803.Methods The cytotoxicity of deguelin (1.56,3.12,6.25 mg/L)and cisplatin (0.78,1.56,3.12,6.25,12.50,25.00 mg/L) in MGC-803 cells was determined by the methyl thiazol tetrazolium (MTT) assay and further confirmed by morphological analysis.Interactions of combination between two drugs were analyzed by isobologram.Cell cycle was assayed by flow cytometry.Results Combination of deguelin and cisplatin exhibited synergistic inhibition of proliferation in MGC-803 cells (P <0.05),induced significant apoptosis and morphological changes.After exposure to deguelin (1.56 mg/L) for 48 h the cells at G1 phase increased.Conclusion Combination of deguelin and cisplatin showed synergistic antitumor effects in MGC-803 cells via cell cycle block at G1 phase.
