Abstract

We have initiated a comprehensive survey of fungi in phase II commercial mushroom compost using sequences of genes encoding small subunit ribosomal RNA (rDNA). A specific segment of 18S rDNA from 26 individually cultured fungi, as well as from total DNA extracted from the compost sample, was amplified using polymerase chain reaction and sequenced. Taxonomic classification of fungal spp. identified by their rDNA sequence was performed by comparison with complete or partial rDNA sequences of previously identified fungi stored in public databases. Corresponding segments of the 18S rDNA of three species of thermophilic fungi, Humicola grisea, H. insolens,and Torula thermophila, were also amplified and sequenced to determine if any of the fungi identified in our compost sample are related to these previously characterized fungi associated with compost. The nature and complexity of the fungal community within mushroom compost is discussed.
