Abstract

The mast cell‐mediated immediate‐type allergic reaction is involved asthma and asthma is a common chronic inflammatory disease regulated by coordination of T‐helper cell type 2 (Th2) cytokines and inflammatory signal molecules. The effect of water extract from Schizandra chinensis Baillon (SCB), known as a traditional folk medicine in Korea, on several cytokines such as interleukin (IL)‐4, IL‐13, tumor necrosis factor‐alpha (TNF‐α), IL‐2 and IL‐3 was investigated in the rat mast cell line (RBL2H3)‐mediated allergy model. Suppressive effects of SCB on Th2 cytokines (IL‐4, IL‐13) was compared with that of digested SCB. The RBL2H3 sensitized with monoclonal anti‐dinitrophenol (DNP) IgE was challenged with the multivalent antigen DNP‐human serum albumin (DNP‐HAS) in the presence or absence of SCB or digested SCB. RT‐PCR and ELISA were performed to measure mRNA and protein expression of cytokines. SCB and digested SCB inhibited the expression of IL‐4 and IL‐13 mRNA and protein in RBL2H3 cells stimulated by IgE‐antigen complex. The result suggested that SCB might suppress the Th2‐mediated response through the down‐regulation of the production of Th2 cytokines.
