Abstract

Membrane geometry and lipid packing actively contribute to the self-organization of reactions at the plasma membrane. However, obtaining quantitative measurements of membrane geometry and lipid packing in living cells remains, indisputably, a non-trivial task. Here, we combine fluorescence lifetime imaging and 3D stimulated emission-depletion microscopy to simultaneously visualize membrane topography (with 2.5+/-0.9 nm axial localization accuracy) and lipid packing at the adherent membrane of living cells.
