Abstract

CGRP constitutes the calcitonin gene-related peptide (CGRP) family along with calcitonin (CT), amylin (AMY), adrenomedullin (AM), and calcitonin receptor-stimulating peptide (CRSP). It consists of a single intramolecular ring structure and an amidated C-terminus, which are common to the family members. CGRP is synthesized and released from sensory nerves in the CNS and gastrointestinal system. It dilates microvasculature in the CNS and is considered to transmit nociceptive signals during migraines. The stimulation of CGRP release is caused by the inflammatory substances in the CNS, and by the activation of the capsaicin receptors in the gastrointestinal system. Cgrp KO mice show high systolic blood pressure and reduced nociceptive hypersensitivity.
