Abstract

Objective To observe the changes of metabolic indexes, left cardiac function of the obese ovariectomized rats after the intervention of Coptidis rhizoma, metformin and probiotics, discuss the possible mechanism of the effects of Coptidis rhizome combined with probiotics on weight, metabolism indexes, left cardiac function of the obese ovariectomized rats. Methods 40 female rats were divided into five groups. After bilateral ovariectomy, the animal model of obese ovariectomized rats were induced by high fat diet. Metformin, probiotics, sterilized deionized water and Coptis combined with probiotics were separately gave by gavage for 6 weeks(sham operation with sterilized deionized water gavage group: abbreviation for HFSS; ovariectomized with sterilized deionized water gavage group: abbreviation for HFOS; ovariectomized with Coptidis and probiotics gavage group: abbreviation for HFOH; ovariectomized with metformin gavage group: abbreviation for HFOM; ovariectomized with probiotics gavage group: abbreviation for HFOB). Weighed all rats’ body weight and Lee index before and after the experiment; detected left cardiac function by bedside echocardiography after intervene. After being sacrificed, heart and other organs of each group were weighed. Results After 4 weeks of high-fat diet, ovariectomized group and sham operation group had a significant difference of weight (P<0.05). Continue the high-fat diet and gavage at the same time for 6 weeks, that’s to say, after 10 weeks of high-fat diet, the weight of all groups increased. HFOH group increased most slowly, HFOM group and HFOB increase slowly and HFOS group is the fastest one. The Lee indexes of five groups increased compared with those of the first week, HFOS group and HFOB group gained the most fast growth. Echocardiography showed that the left cardiac function indexes of HFOH group were significantly lower than other groups such as LVDS and LVESV, the indexes of EF and FS increased significantly compared with other groups. After 12 weeks of high fat feeding, rats were sacrificed to measure their visceral organs which showed heart weight of HFOH group was the lightest. Conclusion Coptidis rhizoma, metformin and probiotics all showed effect of attenuating rapid weight gain and obesity trend in high-fat diet induced obese ovariectomized rats. The intervention of Coptidis rhizoma combined with probiotics showed the most obvious improvement of left cardiac function indexes at the end of experiment and the weight of heart were the minimum.
