Abstract

Optimization of radioimmunoassay (RIA) parameters is essential to obtain a high sensitive and accurate system. The specific activity of the RIA 125I-tracer is a necessary factor to optimize and calculate other RIA factors. A comparative study between two approaches, displacement and graphical, were carried out to calculate specific activity of 125I-antigen of non protein hormone like estradiol (E2). Both direct and indirect of radioiodination of E2 using 125I and tyrosine methyl ester were carried out. Then specific activities of 125I-E2 tracers were calculated by two previous methods. The results show that the displacement calculation more accurate especially in indirect labeling or if 125I-antigen obtained from the commercial market.
