Abstract

1) Nuclei were isolated from early newt embryos using Ficoll in the isolation medium. Nuclei thus isolated were almost free from cytoplasmic contamination, with minimum loss of the nuclear constituents. The characterization of the nuclei thus isolated, including the determination of RNA polymerase activity and analysis of population of RNA's synthesized, is described. 2) In addition, the nuclei isolated from blastulae were characterized by electron microscopic observation, by the determination of RNA polymerase activity and by analysis of the chromatin constituents.
