Abstract

Objective To detect serum biomarkers in prostate cancer(Pca)patients and benign prostatic hyperplasia(BPH)patients by surface enhanced laser desorptjon/ionization-time of flight (SELDI-TOF)mass spectrometry. Methods Forty-five Pca patients and forty-five BPH patients serums were screened by SELDI-TOF mass spectrometry.The data of spectra were analyzed by bioin- formaties tools,Biomarker Wizard. Results Compared with the spectra of BPH patients,there were 9 potential Pca markers detected in the spectra of the Pca patients.In Pca patients,the protein 3771.14,3968.26,4050.39,4279.20,4464.03,4481.59,8919.34 expression levels were higher and the protein 5440.68,6102.82 expression levels were lower than in BPH patients.Using 2 biomarkers (3771.14 and 4481.59)to detect Pca resulted in a sensitivity of 86.7%(39/45)and specificity of 88.9%(40/45).After searching protein database,there were 6 proteins might be potential Pca bio- markers.including Peptide YY,Apolipoprotin C-Ⅱ,Glueagon like peptide(7-37),uncharacterized protein C140rf128,Medin and Neuropeptide Y. Conclusion SELDI-TOF mass spectrometry has great potentiality for exploring Pca tumor biomarkers for early diagnosis and screening.
