Abstract

Objective To detect cardial function in acute myocardial infarction(AMI)rabbits models before and after bone marrow mesenchymal stem cells(BMSCs)with hydrogel composite injection and assess therapeutic effect of this means.Methods Anterior AMI was produced in fifty-six rabbits and BMSCs with hydrogel composite were prepared.One week after AMI,fifty-two rabbits were divided into 4 groups and accepted epicardial injection:①group of treatment(group A),BMSCs with hydrogel composite marked by BrdU;②group of masculine control(group B),BMSCs marked by BrdU;③group of materials(group C),hydrogel marked by BrdU;④group of blank(group D),fetal bovine serum.Echocardiography were performed before AMI,1 week after AMI and 4 weeks after epicardial injection.The conventional echocardiography indices included left ventricular diameter of end diastole(LVDd),thickness of anterior wall(AW)and left ventricular ejection fraction (LVEF).QTVI indices included the peak velocity in systole(Vs)and peak velocity in early diastole(VE).Results Compared with before AMI,rabbits of all 4 groups had significantly larger LVDd.thiner AW,lower LVEF.lower Vs and VE(P<0.05)1 week after AMI.Compared with 1 week after AMI,group A and group B had significantly smaller LVDd,thicker AW,larger LVEF,lager Vs and VE(P<0.05)4 weeks after epicardial injection,which were more significant in group A(P<0.05);but there was no significant difference in group C and group D.Conclusions BMSCs with hydrogel composite can cure AMI effectively,decrease reconstitution of left ventricle and improve local systolic,diastolic function after AMI.
